Specify widths in em's, set to give reasonable line lengths, and let the user chose font sizes that work well with their screen resolution (problem, they probably need exessively small fonts set to cope with all the sites that cram in multiple panels across the width). For most pages, avoid designs which need multiple panels across the width.
