[test-infra] updated README to clarify need for manifest (#1069)

Removed reference to tools/runner, as trying to navigate there through the directory structure rather than directly to the correct file will give a 404.

Clarified that manifest generation is necessary even if you are not creating a new test suite, just to run existing test suites that already have their own manifest file.

Added text on how to get html5lib on an OS X default config where pip is not present.
